AUY922 is a synthetic diarylisoxazole-based small -molecule inhibitor of heat shock protein 90 (Hsp90), that is structurally unrelated to first-generation Hsp inhibitors, geldanamycin and it derivatives. Hsp90 is a molecular chaperone that facilitates the correct folding of specific signal transduction proteins involved in cell proliferation, including human epidermal growth factor receptor-2 (HER2), vascular endothelial growth factor (VEGF), Akt and Raf kinases. Mutant forms of these proteins, as found in some cancers, are more dependent on Hsps. Inhibiting Hsp90 prevents the refolding of key oncogenic proteins, preventing cell proliferation and survival.
