The PROSTVAC vaccine is an off-the-shelf viral vector-based immunotherapy. It utilises two different pox virus vectors which each encode prostate specific antigen (PSA) plus three T-cell costimulatory molecules namely B7.1, ICAM-1 and Lfa-3 (TRICOM). Vaccinia-PSA-TRICOM is replication competent and used for immune priming, while Fowlpox-PSA-TRICOM is a non-replicating virus which is used for repetitive immune boosting.
Monthly subcutaneous injections start with a Vaccinia-PSA-TRICOM priming dose, followed by six, monthly Fowlpox-PSA-TRICOM boosts.
PROSTVAC is currently in Phase II trials for prostate cancer (PC). Previously, two Phase II trials have been completed in this indication. Phase III trials are expected late 2010.
